PurposeTo determine if there is diurnal variation in gene expression in normal healthy conjunctival cells.MethodsBulbar conjunctival swab samples were collected from four healthy subjects in the morning and evening of the same day. The two swab samples were taken from one eye of each participant, with a minimum of five hours gap between the two samples. RNA was extracted and analysed using RNA sequencing (RNA-Seq).ResultsA total of 121 genes were differentially expressed between the morning and the evening conjunctival samples, of which 94 genes were upregulated in the morning, and 27 genes were upregulated in the evening. Many of the genes that were upregulated in the morning were involved in defence, cell turnover and regulation of gene expression, while the genes upregulated in the evening were involved in signalling and mucin production.ConclusionsThis study has identified several genes whose expression changes over the course of the day. Knowledge of diurnal variations of conjunctival gene expression provides an insight into the regulatory status of the healthy eye and provides a baseline for examining changes during ocular surface disease. 相似文献

Understanding the transition from an isotropic liquid state to an organized crystalline state of triacylglycerols has great significance when designing lipid-based materials in general and food products in particular. In the current research, the balance between the demand for the tuning-fork (Tf) conformation, which is the suitable conformation for most crystalline organizations, and the supply of these molecules was discussed. Based on the negative dependency of the nucleation rates and the induction times on temperature, it was suggested that the limiting factor for the nucleation process is the conversion from the Tf conformation to any other conformation. On the other hand, based on the positive dependency of the melting temperatures and enthalpies on crystallization temperature, and the organized morphologies obtained at higher temperatures, it was suggested that the limiting factor during the crystal growth process is the conversion to the Tf conformation from another conformation. 相似文献

Impact of silkworm pupae (Bombyx mori) powder on cream foaming,ice cream properties and palatability
Food fortification with edible insects can offer high nutritional values, improve consumer acceptance and sustainability of resources. This study elucidates the ramifications of silkworm pupae flour (SWF), i.e. dried and pulverized, on the rheological, textural, and sensorial properties of whipping and ice-cream. Results indicate SWF supplementation attenuates cream overrun (p < 0.05) with an observed viscosity increase. Contrary, SWF addition reduced (p < 0.05) separation of foam implying a shelf-life improvement. SWF enriched ice-cream exhibited altered texture attributes (hardness and adhesiveness) especially when using more than 1% (w/w) SWF. In addition, a notable effect on product color was registered which explains the reduced visual appeal of ice-creams containing 2.5% and 4% (w/w) SWF. Moreover, an untrained tasting panel (mean age: 28.6 ± 8.6) reported a strong aftertaste. Overall, this study concludes that SWF at concentrations of ≤1% (w/w) can enhance ice-cream techno-functional properties while maintaining palatability.Industrial relevanceEdible insects are a promising, cost-effective and highly sustainable food resource for proteins and other macro and micro-nutrients. Besides their nutritional advantages, they possess potential techno-functional properties that persist during processing operations and comply with various food matrices. This study offers an original use of bombyx Mori powder, to foam applications, like whipped cream and ice cream. Extraction of plant materials is an essential practice in various fields using natural compounds. Nevertheless, it is challenging to perform complicated extraction efficiently and consistently on materials with hard-to-crush structures. This work presents a new method called simultaneous stone-milling and extraction (SME) to achieve one-step extraction of hard materials. SME under its optimal conditions was confirmed to produce high quality extracts from whole spice seeds (anise seeds, dill seeds, fennel seeds) and coarse-cut star anise. Compared to other extraction methods like maceration, heat-assisted extraction and ultrasound-assisted extraction, SME extracts contained comparable or higher concentrations of general (polyphenols, flavonoids, antioxidants) and specific (enzyme inhibitors) bioactive groups. Hence, SME with its one-step operation is expected to simplify the task of plant extraction and contribute to the advances in studies using plant-derived phytochemicals. 相似文献

AimsConsidering individual variability in regards to the effects of orthokeratology (ortho-k) on myopia progression and controversies regarding the precise underlying mechanism, the aim of this study was to investigate several ocular measurements associated with axial length (AL) growth in children wearing ortho-k lenses.MethodsIn this retrospective chart review, medical records of 53 Chinese children who wore ortho-k lenses over the course of 12 months were reviewed. Baseline variables included age at initiation of ortho-k wear, refractive error (spherical equivalent, SE), central corneal thickness (CCT), and flat and steep keratometry of corneal principal meridians. The change of anterior chamber depth (ACD) and the change of crystalline lens thickness (CLT) between baseline and the 12-month follow-up were also analyzed. The contributions of all analyzed variables to AL change were assessed using univariate and multivariate regression analyses.ResultsInitially, the results of paired t-test showed that CLT and AL were significantly increased after 12 months of ortho-k wear compared with that at baseline (P = 0.001 and < 0.001). The ACD did not change significantly after 12 months compared with that at baseline (P = 0.491). Subsequently, univariate analyses showed that a reduced rate of AL elongation was found in children who were older age at initiation of ortho-k wear (P = 0.028), had greater SE (higher degree of myopia) at baseline (p = 0.006), had thicker CCT at baseline (P = 0.04), and had greater increase of CLT (P = 0.001) in 12 months. At last, only greater SE (higher degree of myopia) and greater increase of CLT were associated with smaller increases of AL in multivariable analyses, (P = 0.003 and 0.001).ConclusionsBoth CLT and AL were significantly increased in children with overnight ortho-k wear after 12 months of follow-up. This work investigated incorporation of Nannochloropsis salina into renneted dairy gels and curd. Whole and ruptured microalgal cells did not impair κ-casein macropeptide cleavage by the rennet enzyme. However, insoluble components of ruptured cells impeded gelation, presumably by hindering interactions between renneted casein micelles. Confocal imaging showed that whole cells were retained and homogenously distributed within the protein network of the gels and cooked curd, whereas ruptured algae formed large aggregates that altered the protein matrix. Eicosapentaenoic acid (EPA) in the whole microalgal cells was incorporated within the curds, with considerably less EPA retained for ruptured cells. Soluble algal debris did not impair gelation, however EPA wasn't retained in the curd. The study demonstrates that nutrient enrichment of renneted dairy products is possible by incorporating whole microalgal cells to displace milk fat with protein and the beneficial long-chain omega-3 fatty acid EPA. Stegobium paniceum (L.) (Coleoptera: Anobiidae) is among the major pests of stored products, causing great damage to stored Chinese medicinal plant materials (CMPMs) in China. Effective control strategies are urgently needed. The aim of this study was to explore the role of volatile organic compounds in the host preference of S. paniceum. First, the olfactory behavioral responses of S. paniceum adults to volatiles from four CMPMs (Panax notoginseng, Angelica sinensis, Gastrodia elata, and Peucedanum praeruptorum) were tested in Y-tube olfactometer experiments. Then, the volatile composition of these plant materials were analyzed by gas chromatography-mass spectrometry (GC-MS). S. paniceum showed significant preferences for volatiles from the four CMPMs, compared with clean air (CA). When S. paniceum was presented with choices among different CMPMs, it showed the strongest preference for P. notoginseng, followed by A. sinensis, then G. elata, and P. praeruptorum. GC-MS analysis identified 43, 34, 28 and 60 components in the volatile profiles of P. notoginseng, A. sinensis, G. elata, and P. praeruptorum, respectively. Falcarinol (14.4%), 3-n-butyl phthalide (78.7%), p-cresol (40.1%), and β-pinene (29.1%) were the most abundant components of the volatiles of P. notoginseng, A. sinensis, G. elata, and P. praeruptorum, respectively. The olfactory responses of S. paniceum to the four CMPMs demonstrated that host-related volatiles play an important role in the host-searching process by adult beetles. This information will be useful for the development of safe and effective trapping strategies for this pest. 相似文献

This study aimed to investigate the effect of limited hydrolysis on conformational and antioxidant properties of soy protein isolate-maltodextrin (SPI-Md) conjugates. Extrinsic fluorescence analysis showed unfolding of the protein molecule and exposure of hydrophobic groups in SPI-Md conjugate hydrolysates. Free amino acid analysis showed that, the contents of hydrophobic amino acids in SPI-Md conjugates increased after hydrolysis. The contents of leucine, isoleucine, phenylalanine increased from 0.32, 0.30 and 0.54 to 1.36, 1.86 and 2.60, respectively, when the hydrolysis degree (DH) gradually increased from 0% to 5.7%. The FT-IR spectrum showed that C = O absorption of the amide group formed by glycosylation continued unabated after limited hydrolysis (DH 2.9%). The glycated SPI products showed good reducing power and superior resistance to lipid oxidation (34%, 12 mg mL?1), whereas the limit hydrolysates (DH 2.9%) of SPI-Md conjugates showed more ef?cient radical-scavenging capacity (89.5%, at 12 mg mL?1) and iron-chelation activity (91.3%, at 12 mg mL?1). Results of this study indicated that, slight enzymatic hydrolysis (DH 0–2.9%) could help partially unfolding the globular structure of SPI-Md conjugates without deteriorating amide bonds and had a positive effect on their antioxidant properties. 相似文献
